چکیده

The present mini-review covers the local and global geometry of framed curves and the computation of twist and writhe in knotted DNA circles. Classical inequalities relating the total amount of bending of a closed space curve and associated knot parameters are also explained.

Knots modify the coil-stretch transition in linear DNA polymers.

We perform single-molecule DNA experiments to investigate the relaxation dynamics of knotted polymers and examine the steady-state behavior of knotted polymers in elongational fields. The occurrence of a knot reduces the relaxation time of a molecule and leads to a shift in the molecule's coil-stretch transition to larger strain rates. We measure chain extension and extension fluctuations as a ...

Research Statement November 2006

Classically, the study of knots and links has proceeded topologically looking for features of knotted curves which depend only on their knot class. Recently there has been an increased interest in geometric knot theory, which attempts to measure geometric properties of a particular knotted curve and relate these to its knot type. Questions about the geometry of knots not only have intrinsic mat...

Many diierent physical systems, e.g. super-coiled DNA molecules, have been successfully modelled as elastic curves, ribbons or rods. We will describe all such systems as framed curves, and will consider problems in which a three dimensional framed curve has an associated energy that is to be minimized subject to the constraint of there being no self-intersection. For closed curves the knot type...

Alexander Modules of Irreducible C-groups

A complete description of the Alexander modules of knotted n-manifolds in the sphere S n+2 , n ≥ 2, and irreducible Hurwitz curves is given. This description is applied to investigate properties of the first homology groups of cyclic coverings of the sphere S n+2 and the projective complex plane CP 2 branched respectively alone knotted n-manifolds and along irreducible Hurwitz (in particular, a...

T7 RNA polymerase cannot transcribe through a highly knotted DNA template.

The ability of T7 RNA polymerase to transcribe a plasmid DNA in vitro in its linear, supercoiled, relaxed and knotted forms was analysed. Similar levels of transcription were found on each template with the exception of plasmids showing varying degrees of knotting (obtained using stoichiometric amounts of yeast topoisomerase II). A purified fraction of knotted DNA with a high number of nodes (c...
